Since there are no detailed regulatory guidelines that direct how stress testing u( u* H+ G Z2 u6 a
is to be done, nor has there ever been a text/reference book on the
a$ @- `" C/ [5 ksubject, stress testing has evolved into an artful science that is highly dependent, |: w Y4 B+ z; V
on the experience of the company or the individuals directing the studies. This# P, w- Z/ V1 O* K* R9 r0 \
book provides readers with a single source reference that benchmarks the
' g( F* A) S' A5 ]3 U. h& K. B* |current best practices of experienced pharmaceutical scientists/researchers.5 z; ?& d0 {3 m9 V; `* k
Questions like ‘‘How hot, how long, what conditions are appropriate?’’ and1 n& Z6 R, _) I- t+ b
topics such as mass balance, photostability, oxidative susceptibility, and the
7 E$ O0 ]/ n9 W" xchemistry of drug degradation are addressed in an objective, detailed scientific$ F$ C- ?$ {* g7 @* A' A1 z
manner, with ample references to relevant guidances and the scientific literature.: F4 ]' a+ t# i' [+ ~3 E3 j
Compiling a book to be used as a scientific reference for the topic of9 r' P8 m) R7 e! m6 m8 c9 j& t
pharmaceutical stress testing was a daunting task. Stress testing, as it relates- d: R" l: D8 _/ N( f& C3 r7 U @
to traditional small molecule pharmaceuticals, is very broad and scientifically2 @$ H- X" r! P/ v5 m+ r6 D9 b
complex. Thus, this book seeks to be a practical and scientific guide: A2 t! L- d" `4 N {
which will, hopefully, stimulate new ideas and further development of the+ X2 M u9 ^+ }, `- F. [) Y' z
science.: ^0 o4 X' y! C
This undertaking is done with some trepidation, since I realize that discussion
' y3 q s0 S1 e5 F! M; r5 xof this topic could lead to attempts to formalize or standardize (i.e.,/ Z4 l8 r) o. [: s3 ]0 B ~6 j3 _& U
regulatory standardization) an area of pharmaceutical science that relies on
7 q9 F2 N2 U' C7 F: s9 O3 _2 Iscientific expertise and flexibility. Each new drug compound, formulation,' L" _5 s! |1 r
route of administration, delivery device, etc. has the potential to present
( g; H5 H# t% V/ Yunique challenges and unanticipated problems, and should be approached
^1 P- R' h2 dwith such a mindset. The pharmaceutical researcher must be free to design
; @# ~6 D" ? j3 M4 Rand develop new ways to predict and measure stability-related issues
. ~0 j% f- Y& H8 B9 R7 l8 N1 [through stress testing. Nonetheless, it is hoped that this book will provide
7 G' C' l$ E9 d. J' ba useful and practical scientific guide that is a welcome addition to the
7 K6 u6 I8 s( `% O- hlibrary of many pharmaceutical scientists. I am confident that the ‘‘artful4 ^: c1 O6 [3 B
science’’ of stress testing will evolve beyond what is represented in this book/ M% p. ?4 S7 k# D2 s& {
as other scientists further the science and fill in topics that are poorly covered
